1. 洗涤缓冲液的制备与储存

手册中明确要求HDL重悬缓冲液需新鲜制备且不可储存。对于1×HDL洗涤液，我个人建议每次新鲜配制，但将其置于4°C储存直至使用是可行的，无需过度担忧。

2. PBS配方建议

在此步骤中，您可根据下游应用需求选择适合的缓冲液进行透析。目前未提及具体PBS配方（如pH、离子强度、是否含Ca²⁺/Mg²⁺或防腐剂如叠氮钠）的强制要求，但需确保其与后续实验兼容。

3. 透析膜选择

对于货号Cat# STA-607，透析方案可采用任意透析方法。例如，可使用赛默飞世尔的Slide-A-Lyzer MINI透析装置。我们通常推荐使用10 kDa截留分子量（MWCO）的透析膜，但其他尺寸也可接受。操作流程需遵循说明书，一般步骤如下：

• 用300倍样本体积的缓冲液透析2-4小时；

• 更换缓冲液，继续透析2-4小时；

• 再次更换缓冲液，过夜透析；

• 建议在4°C下进行透析以保持样本稳定性。

5. 透析后样本情况

• 最终体积：Cat# STA-607的典型产量约为7 ml（来自人血清或血浆样本）；

• 蛋白浓度范围：纯化后HDL的浓度约为4500 µg/ml。

6. 透析后样本处理

需注意，Cat# STA-607提供的溶液并非无菌，因此我们从未对纯化后的HDL进行过过滤除菌。理想情况下，纯化后的脂蛋白可长期储存于4°C；若样本含50%甘油，则可储存于-20°C。关于分装体积，我们无法给出具体建议，需客户根据下游应用自行决定。

7. 蛋白浓度测定方法推荐

手册未明确蛋白浓度测定方法，建议客户根据下游应用选择合适方法。我们通常采用BCA assay测定总蛋白浓度。

8. 总蛋白浓度与HDL浓度的等效性

遗憾的是，我们从未将BCA assay测定的浓度与ELISA结果进行过比较（仅通过ELISA确认ApoB的存在）。因此，无法提供校正方法或参考标准。关于残留的非HDL蛋白，我们通过SDS-PAGE评估纯度。

9. ApoM稳定性

很遗憾，我们从未使用Cat# STA-607评估过ApoM的稳定性（包括其在硫酸葡聚糖沉淀、离心、重悬、洗涤及透析过程中的结合情况），也无相关数据证明其未解离。

10. S1P稳定性

同样，我们从未评估过ApoM或S1P在纯化过程中的稳定性（如离心、涡旋/重悬、透析等步骤是否会导致S1P与ApoM解离或部分丢失）。

11. HDL颗粒完整性

该试剂盒旨在维持HDL颗粒的天然结构（包括脂质双层及载脂蛋白组成），但我们从未对其结构或功能进行过验证（如电子显微镜、天然凝胶电泳等数据）。

12. ApoM-S1P-HDL的沉淀行为

我们从未评估过ApoM或S1P与HDL的结合情况，因此无法确定ApoM-S1P-HDL是否会与总HDL共同沉淀，或是否存在选择性丢失的风险。

13. 最终产物的组成

按照手册操作获得的最终产物是否为“总HDL"？是否包含携带ApoM的HDL亚群（即ApoM阳性HDL）？若有，是否有参考数据（如文献或内部验证）说明其在总HDL中的比例？

很遗憾，我们从未评估过ApoM或S1P的相关情况，无法回答此问题。此外，关于纯化产物中是否残留其他脂蛋白（如LDL、VLDL）或血浆蛋白，我们通过SDS-PAGE评估纯度。

14. 直接兼容性

纯化后的HDL产物（透析后）是否可直接用于ApoM免疫沉淀？是否需要额外的缓冲液交换或纯化步骤（如脱盐、超滤）？

很遗憾，我们从未评估过ApoM或S1P的相关情况，无法给出建议。

15. 残留干扰成分

纯化后的HDL产物中是否有残留成分（如硫酸葡聚糖、Tris、沉淀试剂等）可能干扰抗体-抗原结合？这些残留是否会影响免疫沉淀效率？

通过使用硫酸葡聚糖去除液并进行透析，可去除大部分干扰成分。

16. 抗体推荐

是否有经过验证的ApoM免疫沉淀抗体推荐（如克隆号或目录号）？关于抗体浓度、孵育条件或洗脱方法，是否有建议？

很遗憾，我们从未评估过ApoM或S1P的相关情况，无法提供推荐。

17. 产物组成细节

纯化后的HDL产物中已知成分有哪些（如载脂蛋白种类、脂质组成、缓冲液成分）？是否有批间一致性数据？

我们通过SDS-PAGE评估纯度，但未开展更深入的成分分析或批间一致性研究。

18. 直接纯化产品及技术服务

贵公司是否提供直接纯化ApoM-S1P-HDL的产品（如ApoM免疫沉淀试剂盒、预偶联ApoM抗体 beads/琼脂糖树脂或S1P检测产品）？若没有直接产品，是否有技术建议或定制服务选项用于ApoM-S1P-HDL的纯化？
