A:Sorry for the delay.

Yes, you can mix the lots.  You will have to test, etc for activity.  If endotoxin tests were performed it would be listed on the applicable CoA.  If it is not listed then we did not test for endotoxins.

Q:Hello, may I ask how the extinction coefficient of A137 was obtained? Was it measured experimentally or by other means? It differs quite significantly from the theoretical extinction coefficient based on the sequence. Thank you.

A:Hi Jane,

We have determined FH concentration spectrophotometrically using an extinction coefficient A280 nm (1% solution) of 12.4 (Ref#1). Dr. MK. Pangburn has extensively worked on Factor H. Based on the explanation given by Dr. MK Pangburn in his paper here is the summary: Extinction coefficients reported for factor H have been reported ranging from 9.0 to 19.5 (Ref#1 to 4). The calculated extinction coefficient from the sequence (Ref#4) that has been corrected for the large amount of polysaccharide in factor H is 13.2. Despite these differences, which are typical for a widely studied protein, the value of 12.4 is in the middle of the reported range and is closest to the calculated value after correction for the high content of polysaccharide.
